​Intended Use (IN VITRO DIAGNOSTIC USE)
  • PCRopsis™ RVD Enhancer, when combined with Reagent RVD, Reagent SRVD, or Reagent RVD-RT is intended to enhance extraction-free amplification of RNA and DNA from specimens in compatible transport mediums or saliva.
  • PCRopsis™ RVD Enhancer, when combined with Reagent RVD, Reagent SRVD, or Reagent RVD-RT, may replace nucleic acid extraction procedures for many applications (e.g., life science research, veterinary pathogen research, etc.), but it must be verified by the end user.
​The Science Behind RVD Enhancer
PCRopsis™ RVD Enhancer is composed of a mixture of proprietary peptides, salts, and proteins that synergize with Reagent RVD, Reagent SRVD, or Reagent RVD-RT. RVD Enhancer is particularly helpful in achieving lower Ct values for certain transport mediums and RT-qPCR / qPCR kits. This product is designed to overcome sub-optimal amplification due to secondary structures, nucleic acid protein binding and increased concentrations of PCR inhibitors.

User should verify the applicability of RVD Enhancer for intended applications.

​Product Deterioration
PCRopsis™ ​ RVD Enhancer should not be used if (1) there is evidence of damage or contamination to the product, (2) there is evidence of leakage, (3) the color of the reagent has changed from mostly transparent, (4) the expiration date has passed, or (5) there are other signs of deterioration.​

How to use RVD Enhancer
Optional: Place RVD Enhancer in a 37°C water bath if precipitation or solidification of the product is observed until a clear reagent is observed. This does not affect the functionality of the product.

  1. Thoroughly mix PCRopsis™ Reagent RVD, PCRopsis™ Reagent SRVD, or PCRopsis™ Reagent RVD-RT to ensure homogeneity, but avoid creating bubbles unnecessarily
    1. These products have a hazy, white color when homogenized and normal settlement occurs if not regularly mixed
  2. Mix 2 volumes of RVD Enhancer with 5 volumes of Reagent RVD, Reagent SRVD, or Reagent RVD-RT
    1. Different ratios of RVD Enhancer to Reagent RVD, Reagent SRVD or Reagent RVD-RT can be explored if desired results are not observed
  3. Substitute Reagent RVD, Reagent SRVD, or Reagent RVD-RT with the mixture of RVD Enhancer / Reagent RVD and RVD Enhancer / Reagent SRVD and RVD Enhancer / Reagent RVD-RT in respective protocols

Limitations of the Procedure
  • RVD Enhancer may inhibit qPCR to some extent in certain protocols when using a ratio of 2 volumes of RVD Enhancer with 5 volumes of Reagent RVD, Reagent SRVD or Reagent RVD-RT and the final qPCR mixture consist of ~50% processed sample. No inhibition is observed when the processed sample consist of 25% ~ 35% of the final qPCR mixture. 
  • Repeated freezing and thawing of test specimens may reduce the detection of desired gene targets.
  • Follow recommended guidelines for specimen collection as this may affect the ability to detect desired organisms.

​Performance & CoA
PCRopsis™  RVD Enhancer, when combined with other PCRopsis™ reagents, enables consistent amplification of RNA and DNA gene targets derived from viruses (e.g., coronaviruses) or bacteria (e.g., S. aureus) without the need for nucleic acid extraction procedures. Each lots is carefully tested for sterility and functionality to ensure quality and consistent results.

Storage

This product is ready for use and no further preparation is necessary. The product should be transported and stored in its original container at 25°C until used. Do not overheat. Do not incubate or freeze prior to use. Improper storage will result in a loss of efficacy. Do not use after expiration date, which is clearly indicated on the bottle.

Warnings & Precautions

FOR IN VITRO DIAGNOSTIC USE 
  • Observe approved biohazard precautions and aseptic techniques to prevent contamination of the product. To be used only by adequately trained and qualified personnel.
  • Pathogenic microorganisms, including hepatitis viruses and Human Immunodeficiency Virus, may be present in clinical specimens. "Standard Precautions" and institutional guidelines should be followed in handling all potentially bio-hazardous materials.
  • Sterilize all biohazard waste including specimens, containers and mediums after their use.
  • Directions should be read and followed carefully.
  • Do not re-pack.
  • The use of this product in association with a rapid diagnostic kit, diagnostic instrumentation or used in a manner not intended should be validated by the user.
  • Do not ingest the reagent.
  • Avoid skin contact

Studies with PCRopsis™ RVD Enhancer

Reagent RVD + RVD Enhancer results in lower Ct values when UVT, WHO VTM, CDC VTM and PBS are used as transport mediums

 Various viral transport mediums (BD UVT, WHO VTM, CDC VTM, PBS) were spiked with SARS-CoV-2 and processed with Reagent RVD, without and with RVD Enhancer. The use of RVD Enhancer, in combination with Reagent RVD, resulted in lower Ct values in all viral transport mediums. Reagent RVD alone produced Ct values comparable to extracted RNA when UVT was used as a transport medium. These combined reagents facilitate direct RT-qPCR from RNA viral samples in both UVT and VTM.
RT-qPCR was performed in duplicates using IDT qPCR SARS-CoV-2 probe assay (N1 and N2 gene targets) and Promega GoTaq® Probe 1-Step RT-qPCR System. As a control, RNA was extracted using Qiagen QIAamp Viral RNA Kit.

Satisfaction Guarantee - Warranty Summary

Entopsis guarantees the performance of RVD Enhancer as described in the Certificate of Analysis (CoA). Within 7 days of purchase, contact Entopsis (info@entopsis.com) should RVD Enhancer fail to perform as described in the CoA. In such a scenario, Entopsis will re-test the Lot in question using remaining reagent at our facility at our expense (purchaser does not need to ship product to Entopsis). If the re-tested Lot fails to meet our performance criteria, Entopsis will replace the faulty reagent free of charge. If the Lot meets our performance criteria as stated on the CoA for the Lot in question, then no additional action will be taken.

Please keep in mind that purchasers may use RVD Enhancer for numerous applications, sample types, gene targets, conditions, etc. that are untested and thereby may not work as desired. The purchaser must determine the suitability of the RVD Enhancer for its particular use. However, the Entopsis scientific team can assist in the optimization of RVD Enhancer for your desired application.  Refunds will not be provided due to a lack of product compatibility with purchaser's desired application. This is why small test volumes of RVD Enhancer are offered.
